How to use our Chromatography Campaign Buffer and Consumables Calculator

  1. Choose Planning detail. Select Simple: one buffer for one combined buffer rate, or Detailed: two buffers when each buffer needs its own CV and price.
  2. Enter Packed bed volume (mL) from the packing record or column specification, then enter Planned batches.
  3. In Simple mode, enter Buffer needed per batch (CV) and Buffer cost ($ per L). In Detailed mode, name each buffer and enter its CV requirement and cost.
  4. Open Advanced options only if needed to add Other consumables per batch ($) or a One-time campaign charge ($), then select Calculate.
  5. Sanity-check that Buffer volume per batch matches packed bed volume times total CVs, and confirm that the named buffer volumes match the process worksheet before purchasing.

Definitions

Packed bed volume: The volume occupied by packed resin in the column. This calculator uses it as the basis for all CV calculations.

CV (column volume): A liquid volume equal to one entered packed bed volume. For example, a 1,000 mL packed bed has 1 CV of 1 L.

Buffer volume per batch: The liters of buffer needed for one purification batch from the entered CV requirement.

Buffer cost per batch: Buffer volume for one batch multiplied by its entered cost per liter.

Other consumables per batch: Optional batch-specific spending, such as filters or disposable assemblies, that is added once for each planned batch.

One-time campaign charge: Optional spending added once to the campaign total, without multiplying by batches.

Common mistakes and quick fixes

Mistake: Entering hardware volume or void volume in Packed bed volume (mL).
Fix: Use the packed resin bed volume stated in the packing record or the same volume basis used for the CV requirement.

Mistake: Using a single combined value in Buffer needed per batch (CV) when buffers have different prices.
Fix: Choose Detailed: two buffers and enter separate values for First buffer needed per batch (CV) and Second buffer needed per batch (CV).

Mistake: Entering a total campaign price in Other consumables per batch ($).
Fix: Enter only the cost used for one batch; the calculator multiplies it by Planned batches.

Mistake: Putting a per-batch disposable expense in One-time campaign charge ($).
Fix: Put repeat expenses in Other consumables per batch ($) ; use the one-time field only for a charge added once.

Mistake: Treating Estimated campaign consumables cost as a complete project budget.
Fix: Add any allowed extra charges and separately budget resin, column purchase, equipment, labor, and other items outside this calculator.

Limitations & Key Assumptions / Boundary Conditions

  • The calculation assumes each entered CV is based on the entered packed bed volume. Do not substitute geometric column volume or void volume unless the process worksheet defines CV that way.
  • It estimates liquid demand from entered CVs only. It does not add overage, hold-up volume, line loss, preparation loss, sampling, or spill allowance.
  • Costs use only entered buffer prices, optional per-batch consumables, and an optional one-time charge. Resin, column purchase, equipment, labor, waste, and shipping are excluded unless included in an entered charge.
  • Zero planned batches is valid for checking per-batch demand. Campaign buffer volume and usage-dependent campaign cost will be zero.
  • The calculator does not validate buffer formulation, column loading, recovery, residence time, pressure, cleaning requirements, or manufacturing suitability.

Methodology

Calculation method

One column volume, or CV, is treated as the entered packed bed volume. The calculator converts milliliters to liters using 1,000 mL per L.

buffer volume per batch (L) = packed bed volume (mL) x buffer requirement (CV) / 1000

In Simple mode, the full CV requirement is assigned to one buffer. In Detailed mode, the first and second buffer volumes are calculated separately, then added for the total per batch.

total campaign buffer volume (L) = buffer volume per batch (L) x planned batches

Simple-mode buffer cost is buffer volume per batch multiplied by Buffer cost ($ per L). Detailed mode adds the separately priced first-buffer and second-buffer costs.

estimated consumables cost per batch = buffer cost per batch + other consumables per batch

estimated campaign consumables cost = estimated consumables cost per batch x planned batches + one-time campaign charge

Worked example

With a 1,000 mL packed bed, 5 CV per batch, 10 planned batches, and a buffer cost of $12 per L, one batch needs 5 L. The campaign needs 50 L. Buffer cost is $60 per batch and $600 for the campaign when no optional charges are entered.

Assumptions and limits

Blank optional charges count as $0. A fixed campaign charge is added once, while other consumables per batch are multiplied by planned batches. The estimate uses entered planning values and does not include unentered liquid losses, operational overage, or non-consumable project costs.
