How to use our Cell Seeding Density, Vessel Area and Volume Calculator

  1. Enter the target from your protocol in Target seeding density (cells/cm2) and the actual surface area for one container in Growth area per well or vessel (cm2).
  2. Enter the number of matching containers in Wells or vessels to seed.
  3. Copy the viable, not total, count from your cell counter into Measured viable cell concentration (cells/mL), then enter the complete planned liquid amount in Final volume per well or vessel (mL).
  4. Click Calculate. Use Stock suspension to add per well or vessel only if the Preparation check says the plan is possible.
  5. Sanity-check that Fresh medium to add per well or vessel plus the stock suspension equals the final volume. If the plan is not possible, compare your measured concentration with Minimum viable concentration for this final volume.

Definitions

Target seeding density: The planned number of viable cells placed on each square centimeter of an adherent growth surface, in cells/cm2.

Growth area: The usable surface area inside one well or vessel, in cm2.

Viable cell concentration: The number of living cells in each mL of the measured stock suspension, in cells/mL.

Stock suspension: The cell-containing liquid removed from the measured suspension and added to each well or vessel.

Fresh medium: Culture medium added after the stock suspension so the combined liquid reaches the planned final volume.

Volume room: Final volume minus stock-suspension volume. A negative value means the required stock suspension will not fit in the requested final volume.

Common mistakes and quick fixes

Mistake: Entering cells/mL in Target seeding density (cells/cm2).
Fix: Use the area-based target from the adherent-culture protocol; cells/mL belongs in Measured viable cell concentration (cells/mL).

Mistake: Using total cells in the tube for Measured viable cell concentration (cells/mL).
Fix: Enter the viable concentration reported by the count, in viable cells per mL.

Mistake: Treating Final volume per well or vessel (mL) as extra medium to add.
Fix: Enter the complete final volume after stock suspension and fresh medium are combined.

Mistake: Using a guessed vessel area in Growth area per well or vessel (cm2).
Fix: Check the label or specification for the exact cultureware, then enter its growth area for one well or vessel.

Mistake: Following a negative medium calculation after a failed Preparation check.
Fix: Do not use a negative Fresh medium to add per well or vessel. Increase concentration, increase final volume, or change the density or area.

Mistake: Forgetting to scale the plan for replicates in Wells or vessels to seed.
Fix: Enter every matching vessel that will be seeded, then use Stock suspension needed for the batch and Viable cells needed for the batch.

Limitations & Key Assumptions / Boundary Conditions

  • The calculator assumes every entered well or vessel uses the same target density, growth area, viable cell concentration, and final volume.
  • It uses the viable concentration entered by the user. It does not calculate viability from total cells or a stain result.
  • It does not select a suitable seeding density, volume, medium, or culture condition for a cell line or assay. Follow the relevant protocol or lab SOP.
  • It does not include pipetting overage, dead volume, cell loss during handling, mixing loss, or a safety factor. Add any lab-specific overage separately.
  • A mathematically positive stock or medium volume can still be too small for a lab's reliable pipetting method. Check the equipment and procedure before preparing the mix.
  • The entered growth area must match the exact cultureware. Vessel names and nominal formats can have different areas across products.

Methodology

Calculation steps

The calculator first converts the area-based target into a viable cell count for one well or vessel. It then divides that count by the measured viable concentration to find the stock-suspension volume. Batch quantities multiply the per-vessel quantities by the entered vessel count.

viable cells per vessel = target seeding density (cells/cm2) x growth area per vessel (cm2)

stock suspension per vessel (mL) = viable cells per vessel / viable cell concentration (cells/mL)

fresh medium per vessel (mL) = final volume per vessel (mL) - stock suspension per vessel (mL)

minimum viable concentration (cells/mL) = viable cells per vessel / final volume per vessel (mL)

volume room per vessel (mL) = final volume per vessel (mL) - stock suspension per vessel (mL)

The preparation check is possible when stock suspension is less than or equal to final volume. If stock suspension is larger, fresh medium is not shown as a recipe. The shortfall is the amount by which stock suspension exceeds the final-volume limit.

Worked example

For a target of 10,000 cells/cm2 and a 75 cm2 vessel, the needed count is 750,000 viable cells. With a measured viable concentration of 1,000,000 cells/mL, the stock-suspension volume is 0.75 mL. For a 10 mL final volume, add 9.25 mL fresh medium. With one vessel, the batch totals equal the per-vessel amounts.
